Genetic Fingerprinting of Six Accessions Native Barley in KSA

Abstract

The genetic fingerprinting in six accessions of barley (Hordeum vulgare L.) was investigated by rapid analysis. The accessions were collected from different regions of Saudi Arabia (Al-Taif, Hail, Al-Qassim, Asir, Al-Baha, and Jazan). A total of 91 genetic bands were detected using eight RAPD-PCR primers. The two primers OP-N1 and OP-H1 showed a higher ability to detect high rate of variation between the studied samples .The average variation rate obtained by the eight primers was 92.86%. Cluster analysis based on RAPD-PCR data analysis divided barley samples into two groups, The first group consist of accessions from Al-Baha and Jazan, which shown the highest degree of genetic convergence, and the second group consist of accessions from Asir and Al-Taif, which showed a close genetic relation, while the accessions from Al-Qassim and Hail associated with the second group by a percentage of 57% and 52%, respectively.
