Optimization of camptothecin production and biomass yield from endophytic fungus Fusarium solani strain ATLOY-8

In this study, Fusarium solani strain ATLOY-8, a camptothecin (CPT) producing endophytic fungus was isolated from Chonemorpha fragrans (Moon) Alston. Camptothecin yield from the endophytic fungi was optimized using conventional as well as statistical approaches. Box-Behnken design (BBD) matrix at N=17 was implemented to optimize three independent variables, selected from one factor at a time (OFAT) technique, for maximum CPT production and biomass yield. The BBD exhibited 1.4 and 1.2 fold increase of CPT production and biomass yield respectively compared to OFAT approach in the presence of the basal medium containing absolute ethanol (5%), glucose (1%) and precursors (combination of tryptophan, geraniol and tryptamine; 0.03%). Analysis of variance (ANOVA) revealed high coefficient of determination (R2) of 0.9751 and 0.9980 respectively for CPT and biomass yield at significant level (P>0.05). Three dimensional graphs revealed interdependent interaction between two variables in order to improve the CPT and biomass yield.
