In this study, callus induction trait in different
tomato genotypes were detected through three
categories of genetically significant markers,
morphological markers, protein-based markers
and DNA- based markers (RAPD). Both
molecular and biochemical markers associated
with this trait offered a promising alternative to
the morphological markers as they can be
analyzed at a very early stage of plant
development and mostly display dominant
inheritance. Thus, complete information about
this trait can be obtained in a very early stage
to decide the appropriate selection and
crosses required for production of plants with
good callus induction ability to be used in
many applications.
Moreover, the in vitro performance of diverse
groups of tomato cultivars and the mode of
inheritance of callus induction were examined.
In addition, the effects of many recommended
culture media recipes for callus induction
ability were measured. SDS-PAGE and RAPDPCR
techniques were used to find out any
biochemical and molecular markers associated
with trait respectively. It worth to be mentioned
that some molecular and biochemical markers
were revealed which could be used in marker
assisted selection (MAS).
