PHYLOGENETIC RELATIONSHIPS FOR SOME TILAPIA SPECIES USING ELECTROPHORESIS

Abstract

Phylogenetic relationships of tilapia fishes collected from different locations in Egypt, namely Maryout, Manzala and Abbassa farm (CLAR) in Sharkia, Egypt were examined. RAPD-PCR technique and SDS-PAGE for muscle protein were used.
Six randomly primers (OPA-5, OPB-17, 18, 20, OPC-02 and OPD-12) were tested and bulked samples were subjected from each population and produced distinctive bands. Genetic similarity estimation revealed that Oreochromis niloticus (Maryout) and (Manzala) were very close and showed high similarity (0.92) followed by O. aureus (0.62) and T. zillii, which was distal (0.47). Moreover, the complexity of DNA fingerprinting varied from one species to another, suggesting that the bulked sample can be particularly useful in breeding programs and hybridization.
SDS-PAGE for eye protein of O. niloticus (MR), showed total numbers of 28 bands, while O. niloticus (MZ) had 30 bands with different relative mobilities. O. aureus and T. zillii showed 33 and 25 bands with different relative mobilities, respectively.
The results generated from the RAPD and protein analysis were pooled for drawing the genetic relationships among the examined populations. The dendrogram indicated that O. niloticus of Maryout and Manzala form one group. O. aureus was close to O. niloticus than T. zillii.
